Selection and Validation of miRNA Reference Genes by Quantitative Real-Time PCR Analysis in Paeonia suffruticosa

The miRNA, a kind of endogenous non-coding small RNA, plays an essential role in regulation gene expression plants. Quantitative real-time PCR (qRT-PCR) assay is one the most common methods used for quantification miRNA expression, and levels are normalized by comparing with reference genes. present study was intended to identify appropriate genes normalizing level various developmental stages tissues such as bud development process, flower stages, different tree peony flowering times. Five algorithms (Delta Ct, geNorm, NormFinder, BestKeeper, RefFinder) were stability analysis. results showed that mtr-MIR160b-p3 gma-miR394a-5p stable miRNAs expressed during process early-flowering ‘Feng dan’; PC-5p-19095 late-flowering ‘Lian he’, followed mtr-MIR160b-p3; mtr-miR159a varieties. PC-3p-871 dan’, PC-5p-4, PC-5p-4 mtr-miR168b. findings this provide studying changes further exploring regulatory mechanism peony.
